What is tert-Butyldimethylsilyl Chloride?
White solid. Melting point 86-89℃, boiling point 128℃. Soluble in most organic solvents, usually used in CH2Cl2, THF and DMF. The reagent is sensitive to moisture and has a pungent taste. This product is corrosive and flammable. It is recommended to be used in anhydrous systems and operated and used in a fume hood.
Tert-butyldimethylchlorosilane (TBDMSCl, TBSCl for short) is an important sterically hindered organosilicon protective agent. The active hydrogen in organic compounds can be replaced by silyl groups under mild reaction conditions to generate intermediates with high chemical stability, and the reactivity of other groups is not affected. The role of TBDMSCl in reacting with alcohol hydroxyl groups to generate silyl ethers is particularly important.
Compared with the common trimethylchlorosilane protective agent, TBDMSCl silyl ether has higher stability. Due to its steric hindrance, it has excellent selectivity on polyhydroxy substrates and can easily selectively protect primary alcohols in the presence of secondary or tertiary alcohols. Tert-butyldimethylchlorosilane (TBDMSCl) is a commonly used silanizing agent and one of the most widely used important protecting groups in organic synthesis. It is used as a protecting group for hydroxyl groups in the synthesis of ribonucleosides, and is also an oxidant and decyanant. It can react with hydroxyl groups under mild conditions to form the corresponding silyl ethers, and react with ketones, esters or amides to form the corresponding enol silyl ethers.
