Product Introduction:

Tris(2-carboxyethyl)phosphine hydrochloride (TCEP hydrochloride, TCEP HCL) is a novel reducing and deprotecting agent for thiolated DNA. Its products are the best among trivalent phosphorus derivatives. It is a colorless, odorless, weakly reducing substance soluble in water, and its solubility in water can reach 310mg/mL.

TCEP HCL has high stability in air and aqueous solution, and is less affected by temperature and concentration. It has mild reactivity, easy solubility, low toxicity and easier operation. The stability of TCEP-HCL in acidic and alkaline solutions is very good.

TCEP is a very suitable disulfide bond reducing reagent, which is extremely selective for reducing disulfide bonds. Except for cysteine, there are almost no side reactions with other amino acids. And it can be used in a wider pH range including acidic conditions (pH value 3), thereby effectively reducing the hydrolysis of amide bonds.

The reducing properties of TCEP HCL compared to DTT:

The reducing ability of TCEP HCL is stronger than that of DTT, and it is more stable than DTT. Compared with DTT, TCEP hydrochloride can reduce disulfide bonds in a wider pH range. Its effect can be maintained for 2 to 3 weeks and is recognized as a good substitute for DTT (dithiothreitol).

Used as water-soluble reagent:

Tris(2-carbonylethyl)phosphorus hydrochloride is an organophosphorus compound that can be used as a water-soluble reagent. For the selective reduction of disulfides, it can be used to prevent intermolecular or intramolecular disulfide bonds created between cysteines in proteins.

Used as reducing agent:

TCEP HCL is a non-thiol reducing agent that is more stable than other chemical reducing agents and produces a faster S-S reduction reaction. Much more steady than DTT in mass spectrometry applications. It can also be made use of for discerning decrease of disulfides in water.

Because TCEP hydrochloric acid does not contain a sulfur structure, its reduction reaction solution does not require additional purification, and can be directly used in solutions containing maleimide or cross-linking reagents. In general, TCEP HCL concentration less than 10-20mMol/L can be directly used in chemical reactions containing maleimide.
