What is Levan?

Fructans are linear and/or branched polymers composed of fructose, and are important storage carbohydrates for about 15% of angiosperms (Hendry, 1993). Research on fructans in various countries around the world can be traced back to 1804, and has a history of about 200 years (Rose, 1804). Early research mainly focused on the physiological and biochemical properties of fructans. With the discovery of fructan metabolic enzymes, the focus of research shifted to the understanding of fructan metabolic processes and functions (Pol-lock and Cains, 1991).

Levan is now found that in addition to being a short-term or long-term storage material for heterotrophic plant organs – roots, stems, seeds and autotrophic organs – leaves, fructans also have a variety of important physiological functions, such as regulating plant adaptation to low temperature photosynthesis (Koops and Jonker, 1996; Pollock, 1986), regulating plant sucrose distribution (Pollock and Cains, 1991), and adapting plants to water deficit (De Roover et al, 1999; Hincha et al, 2000; Pilon_Smits, 1995), high CO2 concentration (Smart et al, 1994), and hypoxic conditions (Albrecht et al, 1993).

In addition, fructans have become important health products due to their high sweetness and low caloric value, as edible sweeteners and low caloric food raw materials. Generally speaking, low DP (Degree of polymerization, number of fructosyl groups) fructans are not easily digested, can prevent tooth decay and weight gain, and can correct intestinal flora imbalance, etc. High DP fructans have excellent physical and chemical properties and can be used as nutritional foods (to replace fat) and non-food raw materials (Fuchs, 1993).

Levan Uses

Levan have a wide range of industrial applications in the fields of cosmetics, medicine or nanotechnology. It may have anti-hyperglycemic and anti-tumor effects, prevent diabetes-induced oxidative stress, and increase catalase (CAT) and superoxide dismutase (SOD) in the heart. Fructans are also recognized for their immunomodulatory and prebiotic effects.

Levan from Erwinia herbacea are used as:

  • Nuclear magnetic resonance (NMR) spectroscopic standards for fructans produced by Bacillus licheniformis NS032 in beet molasses medium.
  • Polysaccharide substrates to test the ability of Prevotella isolates to grow in vitro on polysaccharides as the sole carbon source.
  • Substrates in fructanase activity assays.

Physiological Functions

  1. Short-term storage of sugars.
  2. Regulation of the size of sucrose pools in photosynthetic tissues.
  3. Maintaining appropriate water potential in plant tissues through osmotic regulation.
  4. After depolymerization into fructose molecules, the freezing point of water in the tissues is lowered, increasing the cold resistance of plants.
  5. Enable plants to photosynthesize at low temperatures. It is known that the photosynthetic rate of plants that accumulate starch is greatly reduced at low temperatures. However, the photosynthetic rate of plants that accumulate fructans does not decrease at low temperatures, even at 0. C, so the plants can grow normally.

Classification of Fructans in Higher Plants

Fructans are polymers formed by connecting sucrose with one or more fructose molecules, and are mainly found in bacteria and higher plants. Fructans in bacteria are timothy sugar-type fructans, which are extracellular polysaccharides and are 2 to 3 times larger than fructans in higher plants. Fructans in higher plants have a very complex structure, and their chain length varies greatly. They are generally composed of 30 to 50 fructosyl units, and sometimes the DPs (fructosyl number, also known as degree of polymerization) is greater than 200. The shortest fructan is fructotriose (cane fructotriose). So far, the following 5 main types of fructans have been found in higher plants.

  1. Linear inulin-type fructan: It is formed by connecting one or more furanoses with 1-kestose as the basic unit through β(2→1) bonds. The shortest linear inulin-type fructan is 1-kestose or isokestose, which is formed by connecting fructose to the 1 carbon position of the fructosyl group in the sucrose molecule. Linear inulin-type fructan is unique to Asteraceae plants, such as Jerusalem artichoke and chicory.
  2. Linear inulin-type fructan neoseries: It is formed by connecting the fructosyl units connected by linear β(2→1) bonds to the C1 and C6 positions of the glucose part of sucrose, that is, a fructan polymer with fructose chains at both ends of the glucose molecule. The smallest linear inulin-type fructan neoseries molecule is a new kestose formed by connecting a fructosyl group to the 6 carbon position of the glucose group in the sucrose molecule. , both ends of its glucosyl group are fructose. The new series of linear inulin-type fructans are mostly found in lily plants such as onions and reeds.
  3. Linear levan-type fructan: 6-kestose is used as the basic unit, and one or more fructose are connected by β (2→6) bonds. The smallest levan-type fructan is 6-kestose. Linear levan-type fructan exists in some grass plants, such as Dactylis glomeratus and Phleumpratense.
  4. Mixed levan: It is composed of levan as the basic unit, with one or more fructose connected at either end by β(2→1) and β(2→6) bonds. Mixed levan exists in many higher plants, such as wheat (T.aestivum), barley (Hordeumvulgare), etc.
  5. Levanneoseries: It is a polymer formed by connecting fructosyl units to both ends of sucrose molecules by β(2→1) and β(2→6) bonds. This type of levan is only found in a few plants such as oats (Avenasativa).
