When a regulatory dossier for an infant formula marketed in the European Union arrived at a notified body for review in 2023, the HMO ingredient list included both 2′-fucosyllactose and lacto-N-neotetraose. The reviewer’s first question to the manufacturer was not about efficacy data. It was about the structural characterization package for LNnT — specifically, whether the submitted HPLC method was capable of resolving LNnT from its type-1 chain isomer, Lacto-N-Tetraose. The two tetrasaccharides differ only in the linkage position connecting the internal galactose to the N-acetylglucosamine residue: type-2 chain in LNnT (β1-4) versus type-1 chain in LNT (β1-3). Most standard reversed-phase HPLC methods cannot differentiate them without optimized conditions, and the manufacturer was required to submit supplemental NMR data before the dossier could advance.
